Genetic Diversity Analysis and Screening of High Psoralen Yielding Chemotype of Psoralea corylifolia from Different Regions of India Employing HPLC and RAPD Marker

Psoralea corylifolia, an important medicinal herb of Fabaceae is a rich source of secondary metabolites, furanocoumarins (psoralen, isopsoralen) and isoflavonoids (genistein, daidzein). A wide variation in terms of genetic diversity and psoralen content has been recorded through RAPD and HPLC analysis, respectively among various chemotypes of P. corylifolia collected from eight different regions of India, viz. Ghaziabad, Haridwar, Khari- Baoli, Kolhapur, Mansur, Manesar, Sirohi and Sirsa. A high degree of polymorphism (89.68%) was revealed among the eight chemotypes studied. The highest genetic similarity co-efficient (0.767) was observed between the chemotype accessions of Sirsa (Haryana) and Khari Baoli (Delhi), while the lowest value of 0.512 was revealed between two groups proving that there is a wide diversity among different chemotypes. HPLC analysis of the seeds of the aforesaid chemotypes revealed that chemotype of Sirohi region yielded the optimum content (4763.00 μg/ g fresh wt.) of psoralen over the others. This is our first report which provides scientific basis for authentication of relatively high diversity in terms of genetic and biochemical constitution among different chemotypes of P. corylifolia collected from various regions of India for detection of elite germplasm for commercial purpose..
