Abstract


Staining procedures are designed to improve the observation of cells that allows detecting and tracking any type of pathology under the microscope. Several techniques can be performed according to the purpose of the examination, type of material and the specimen obtaining method, as they are determinant in the choice of staining technique. The aim of this study was to describe Papanicolaou technique preparation of dye solutions methods and some staining protocols in order to aid in the diagnosis. Research has been done from manuals, database, staining techniques books, inspection of colouring cytological and histological routine and later took place experiments by changing the pigment concentration, for assessment the final result on the colour appearance. Our results revealed that a surplus amount of yellowish eosin or green light, of the EA original formulation, caused a solution unbalance, producing different results to what was expected of a equalized solution. Follow the training manuals as standard procedure associated with a constant monitoring and strict quality control, characterizes as the main tools to avoid flaws which could compromise the lamina reading, ensuring the examination excellence and the results quality.
